SPRi-Biochip functionalized with NHS group at low density
CS-LD surface chemistry is suitable for immobilizing molecules with an amine function (NH2 group). The immobilization principle is based on the high affinity of sulphur atoms in POE molecules (POE = polyoxyethylene) on gold.
Free carboxylic acid functions of POE are then activated by a NHS:EDC solution (NHS = N-Hydroxysuccinimide, EDC = 1-ethyl-3- (3-dimethylaminopropyl) carbodiimide). Activated carboxylic acids can then react with free amine functions of the ligand to create amide functions.
CS-LD chemistry (CS surface chemistry low density) is used to immobilize molecules with high molecular weight.
