How the Sample Ref Accessory compensates for environmental changes in Raman analysis

In Raman spectroscopy, the accuracy of measurements does not depend solely on the sample being analyzed. Variations in the measurement environment, such as changes in ambient temperature or drift in the laser and spectrometer, can also influence the data obtained. These variations can become particularly problematic when it is necessary to accurately measure small shifts in Raman bands.

To limit the impact of these phenomena and improve measurement reliability, a reference sample can be measured simultaneously with the sample under study. This is precisely the principle behind the Sample-Ref Accessory, a reference accessory for monitoring environmental changes in Raman microscopy.

Why can changes in the environment affect Raman measurements?

Raman analysis relies on the precise measurement of the position and intensity of spectral bands. When measurement conditions change during an experiment, the variations observed in the spectrum do not necessarily originate from the sample itself.

For example, a change in room temperature can alter experimental conditions and affect the measured spectral position. Similarly, drift in the laser or spectrometer can cause a shift in the data over time. The Sample-Ref corrects data affected by changes in ambient temperature or laser drift.

This issue becomes particularly important when the goal is to measure very small shifts in wavenumber. In applications such as the study of stress and strain in materials or the analysis of graphene, a few hundredths of a cm⁻¹ can be significant. Controlling spectral stability then becomes essential for correctly interpreting the results.

A reference measure for identifying and correcting deviations

The principle behind the Sample-Ref Accessory is to record the spectrum of a reference sample simultaneously with that of the sample being analyzed. This reference measurement provides a point of comparison that allows monitoring of any system variations during the analysis.

This approach can be used, in particular, to correct spectral drift in the spectrometer. A neon lamp can be recorded alongside the sample’s spectrum to track this drift. The simultaneous use of a reference sample also makes it possible to correct drifts in the laser and the spectrometer.

The benefit, therefore, is that each measured variation is no longer necessarily attributed to the sample. The reference allows tracking changes that affect the measurement system and taking them into account during data processing.

Also monitor changes in laser intensity

Changes in the environment are not the only factors that can cause results to vary. Laser power can also fluctuate during an experiment, leading to changes in the measured Raman intensity.

The Sample-Ref allows for the simultaneous measurement of a reference sample and the sample of interest in order to monitor these intensity variations and correct them.
This approach can be particularly useful when measurements extend over time or when comparing spectra requires stable experimental conditions.

A solution tailored for high-precision Raman measurements

The Sample-Ref Accessory can be mounted on the LabRAM HR Evolution’s upright open-space microscope (FSM).

On a LabRAM HR Evolution equipped with this accessory, very high spectral precision can be achieved, typically better than 0.02 cm⁻¹. This feature is particularly valuable for applications requiring precise tracking of small shifts in Raman bands.

Ensuring the reliability of measurements by controlling system variations

In Raman spectroscopy, improving data quality is not just a matter of optimizing the sample or the acquisition parameters. System stability and the ability to identify variations unrelated to the sample are also important.

By incorporating a simultaneous reference measurement, the Sample-Ref Accessory provides a way to monitor drifts that could affect results: temperature variations, laser drift, spectral drift of the system, or intensity variations. The data can thus be corrected to account for these effects, enabling a more reliable interpretation of small spectral shifts.
For Raman applications requiring high wavenumber precision, particularly in the study of stress and strain or graphene, this approach therefore allows for better differentiation between variations related to the instrumental environment and the phenomena actually observed on the sample.

In summary, the Sample-Ref Accessory does not eliminate environmental variations; rather, it allows them to be monitored, characterized, and their impact on the Raman data to be corrected. This approach is particularly relevant when spectral precision and the comparability of measurements are essential.
